Vector

Vector is a DNA molecule used as a vehicle to transfer foreign genetic material into another cell. Vector introduces recombinant DNA into the cell plant.  It also amplifies and  sustains within the cell. The term `vector` is derived from the Latin word ‘vehere`.

In the experiment to be conducted in GenoCon, Vector is a vehicle which carries your designed DNA sequences into chromosomes of Arabidopsis thaliana.

Firstly, DNA sequences are to be synthesized based on the selected designed DNA sequences by Document Review. Secondly, the DNA sequences are introduced into Arabidopsis thaliana by vector named pMDC32. A promoter and a terminator are already set in the vector. Please make use of those in your DNA design.

35S promoter: cauliflower mosaic virus (CaMv) 35S promoter. `2X` expresses two promoters in a line which means more powerful than a 35S promoter.

Nos terminator: terminator which derives from nopaline synthase (NOS) genes of a bacterium named Agrobacterium  tumefaciens

attR1, attR2: recombination site derived from λ Phage in the Gateway® 5 cloning system. (The designed DNA sequences are inserted into the site between attR1 and attR2.)

LB: Boundary area in the left side of T-DNA (Far left part of the sequences which are to be introduced into chromosome of Arabidopsis thaliana.)

RB: Boundary area in the right side of T-DNA (Far right part of the sequences which are to be introduced into chromosome of Arabidopsis thaliana.)

Black bar: sites for various restriction enzyme are set (We do not use them for the experiment in GenoCon.)

Reminders upon designing DNA sequences

The designed DNA sequences are inserted into a site between attR1 and attR2. As you see in the picture above, DNA sequences between LB and RB are to be introduced into the test plant, please keep it in your mind that 35s promoter and Nos terminator are to be added to your designed sequences by use of pMDC32.
